DetectX^®硫醇检测试剂盒用于定量测量生物样品中产生或存在的硫醇基团。
提供标准品以生成测定的标准曲线，并且从生成的曲线中读取所有样品。将样品和标准品吸移到黑色微孔板中，将样品或标准品与ThioStar混合，并在室温下孵育30分钟后，在荧光板读数器中以510 nm的波长在390 nm处读取荧光产物。使用大多数荧光板读数器提供的软件对样品稀释液进行适当校正后，可以计算出样品中硫醇的浓度。试剂盒提供了一个用于测量的96孔板，但该测定法适用于更高密度板格式的测量。用户在运行样品之前，应确保其黑色板适合与这些试剂一起使用。由于生物产生的游离巯基，例如谷胱甘肽和蛋白巯基存在于不同的环境中，因此我们建议最终用户使用合适的标准品校准存在或产生的硫醇的量。

Background: Free thiols in biological systems have important roles. Oxidatively-modified thiol groups of cysteine residues are known to modulate the activity of a growing number of proteins. As such, it is important to be able to accurately determine the extent of modification of specific amino acids, such as cysteine residues. This is especially difficult in a complex protein sample, especially in the presence of chaotropic agents such as guanidine hydrochloride. Typical methods using Ellman’s reagent do not have sufficient sensitivity to allow economical detection of free SH groups.
